We are currently searching for a Scientist to Investigate the pathways of genome metabolism involving helicase enzymes defective in hereditary disorders displaying age-associated clinical features and/or cancer. Understand the molecular and cellular roles of human DNA helicases in pathways that are important for the maintenance of genome integrity and cellular homeostasis. Genetic and biochemical approaches are used to characterize the roles of helicases in cellular nucleic acid metabolism through their catalytic functions and protein interactions. Use of siRNA/CRISPR screens, pharmacologically active small molecules, single-molecule DNA fiber studies, live cell imaging by immunofluorescence confocal microscopy, protein biochemistry, and enzyme kinetic assays.

This opportunity is full-time, and it is on site in Bethesda, MD.

Duties & Responsibilities
  • Perform investigations with reagents material/ tissues under investigation which range from cellular products to whole tissue; maintain inventory system and sample storage; maintain inventory of all experiments.
  • Perform biochemical assays including electrophoretic mobility shift assay EMSA, helicase assays with radiometric or fluorometric substrates, ATPase assays, protein interaction assays ELISA, co-immunoprecipitation, dot blot, affinity pull-down.
  • Present ongoing research and findings at weekly lab unit section meetings.
  • Perform mathematical calculations for the preparation of graphs.
  • Write and edit scientific papers for submission to peer-reviewed journals.
  • Present papers/abstracts/posters at scientific meetings/conferences.
  • Design and synthesize nucleic acid substrates for biochemical assays to analyze proteins involved in nucleic acid metabolism.
  • Work with staff on the maintenance of animal protocols; work with animal facility staff to maintain, design and produce the necessary crosses of distinct mouse strains Safeguard animal logs; work with animal facility staff to maintain and breed specific mouse models.
  • Work with staff on the maintenance of laboratory protocols; work with facility staff to maintain, design and produce the necessary safety protocols and procedures.
  • Perform molecular biology experimentations, bioinformatics, gene and protein profiling, protein concentrations in extracts.
  • Express recombinant proteins using a baculovirus system and purify recombinant proteins using both conventional biochemical and chromatography steps, as well as design protocols and operate FPLC instrumentation.
  • Work with staff on scheduling and maintenance of research tracking system and databases for all research activities, daily records of experiments and results, including research documentation.
  • Maintain an inventory system and sample storage per outlined guidelines by making la-bels, tubes, aliquoting and re-inventory of processed samples; ensure that policies and procedures regarding retrieval are followed Maintain an inventory and notebooks of all experiments.
  • Create and maintain a research tracking system and database for all research activities, daily records of experiments and results, including research documentation.
  • Keep up to date on technical developments, especially in the fields of genomic stability, DNA repair, DNA damage response, cellular senescence, nucleic acid metabolism, nucleic acid processing enzymes and binding proteins, recombinant protein expression and purification, single-molecule studies, and single-cell experimentation.
  • Operate laboratory equipment to analyze samples.
  • Use or operate a variety of routine and specialized laboratory instruments and equipment for the collection of experimental data from various molecular, cellular, tissue, and organismal assessments, and analysis of data.

Requirements

Qualifications:

  • Minimum Seven (7) years of experience in Molecular Biology, Protein Biochemistry, or a related field.
  • PhD degree in Biological Sciences or other related discipline.
    • OR Master's degree plus three (3) years of specialized
    • OR BA/BS degree plus five (5) years of specializedexperience


Skills:
  • Knowledge of proper laboratory disposal methods and safety procedures.
  • Ability to work independently and think critically with skilled hands and sound
  • Strong communications skills, both oral and written.
  • Excellent analytical, organizational and time management skills.
  • Experience with isolation of DNA and RNA from tissues and cells, small samples, PCR, immunohistochemistry, Western blotting, current methodologies of gene targeting and analysis; RNA interference, transfections, and prior experience working with cell models.
  • Familiarity with flow cytometric techniques.
  • Familiarity with tissue culture techniques.
  • Strong familiarity with cell biology, genetics, state-of-the-art molecular biology, and its techniques.
  • In-depth knowledge of single-cell DNA replication and DNA repair assays and single-molecule (kinetic) biochemical assays.
  • Experienced with MS Office.
