OPPORTUNITY
Mammoth is seeking an Engineer III with protein production expertise to join its growing Discovery team. The Discovery team is responsible for developing novel CRISPR-associated (Cas) nucleases for genome editing applications.

The successful candidate will be responsible for executing laboratory workflows to develop Cas nucleases for genome editing. The primary focus for this role will be on protein production and process development. The successful candidate will also have the opportunity to contribute broadly across technical areas, including plasmid construction, NGS assays, and high-throughput screening. KEY RESPONSIBILITIES
  • Manage internal and external research-grade protein production to support Discovery and Preclinical research activities
  • Execute internal protein production
  • Execute process development to establish methods to produce proteins and protein complexes for biochemical characterization and structural interrogation
  • Rigorously document newly developed methods and results
  • Provide regular updates at internal meetings
  • Other duties as assigned

REQUIRED QUALIFICATIONS
  • Bachelor's degree in molecular biology, biochemistry, or a related field
  • 5+ years of laboratory experience developing and executing bacterial protein expression workflows including, but not limited to:
  • Downstream protein process development including target protein extraction, resin scouting (ion-exchange, affinity, size exclusion), phase-partitioning, filtration, etc
  • Protein analytical methods (Eg. homogeneity evaluation, protein quantitation, detection / quantitation of process-related impurities such as host-cell protein, endotoxin, nucleic acids)
  • Generation and proper documentation of master and working cell banks
  • FPLC systems (Eg. Cytiva AKTA, BioRad NGC)

PREFERRED QUALIFICATIONS
  • Prior experience managing external partnerships (Eg. CRO, CMO)
  • Previous protein manufacturing experience (research-grade or cGMP)
  • Hands-on experience in the following areas: biochemical assays; molecular biology (PCR, DNA/RNA purification and quantitation)
  • Computer and scripting skills (Benchling, Geneious, JMP, SQL, R, python)

TEAM
Our small-size proteins, such as Cas14 and CasPhi, are poised to transform disease treatment enabling novel editing approaches and delivery options that are difficult to achieve or not even possible with previously described CRISPR-Cas systems. Our teams are responsible for discovering and engineering novel CRISPR systems, establishing CRISPR-based diagnostics, and translating our unique CRISPR technology into therapeutic treatments with the ultimate goal of cures for indications with high medical need.
