This position is responsible for leading the optimization and scale up of processes, troubleshooting manufacturing issues, improving product quality, and reducing manufacturing costs. The candidate will be responsible for conducting research and manufacturing related to vector development, fermentation, protein expression, protein purification, protein characterization, and assay development. The position will be required to work independently at the bench as well as effectively collaborate through communication of scientific findings to internal and external groups. The job function requires sound scientific judgment and innovation using advanced practices and procedures to achieve solutions.

Relationships:
  • Key Stakeholders
  • This position will collaborate with Technical Operations and Development to design experiments, execute experiments, and analyze generated data for assigned projects. The position will collaborate with various groups including quality and operations to complete both development projects and assigned custom manufacturing.
  • This position will interact with both internal and external contacts. This position will present scientific findings to the appropriate managerial teams. This position will be responsible to communicating with contract manufacturing clients regarding progress and technical issues.
  • Line Management
  • This position reports to the Manager, Technical Operations and Development.
  • This position can be an independent contributor or manage Senior Research Associates and Research Associates as assigned.
  • The direct reports for this position, if applicable, will be individual contributors.
Key Duties
  • Collaborate within multidisciplinary development and operational teams to improve and optimize products and processes, scale up processes, and manufacture custom products.
  • Design, clone, and optimize constructs that will be used to express proteins in Escherichia coli , Saccharomyces cerevisiae , and Pichia pastoris for commercial manufacturing.
  • Optimize and scale-up protein expression through research to understand fermentation parameters and media optimization.
  • Characterize microbial cell lines and microbial cell banking including growth, expression, and stability.
  • Develop assays that characterize protein expression and protein quality early in the production workflow.
  • Develop, execute, and scale up protein purification processes for recombinant proteins, monoclonal antibodies, and polyclonal antibodies. Unit operations include, but are not limited to, preparative centrifugation, depth filtration, tangential flow filtration, chromatographic methods (ion exchange, size exclusion, affinity, etc.), and product formulation.
  • Develop and perform various bio-analytical methods for process monitoring and product characterization. The methods include, but are not limited to, SDS-PAGE, Western Blot, ELISA, spectroscopic methods, HPLC methods.
  • Document and manage experimental methods and results within laboratory notebooks or manufacturing documentation. Prepare routine technical progress reports to be shared at internal and external meetings.
  • Help identify new technologies for protein and virus bioprocessing and characterization.
  • Lead technical transfer of developed procedures to production and/or quality personnel. Includes development of batch records, design of transfer plan, and execution of transfer plan including boots on the ground training.
