OPPORTUNITY
Mammoth Biosciences is seeking a Sr. Scientist I with expertise in high throughput molecular biology and strain construction for its Diagnostic Protein Engineering team. This role will focus on using high throughput molecular biology methods for generating optimized proteins for next-generation CRISPR diagnostic assays.

A successful candidate in this position will be responsible for proposing and executing strategies for high throughput generation of CRISPR Cas nuclease variants for diagnostic applications. This role will function as part of the Assay Innovation group, which is responsible for developing new diagnostic technologies and sits upstream of our Assay Development group.
KEY RESPONSIBILITIES
  • High-throughput molecular cloning and diversity generation for protein engineering targets (site saturation mutagenesis, domain swapping, combinatorial variant construction, etc.)
  • Bacterial growth and protein expression optimization (including consideration of strain/promoter optimization)
  • Designing and performing Next-Generation Sequencing (NGS) assays
  • Analysis of large datasets
  • Document experimental results in laboratory notebooks
  • Present experimental results at internal meetings
  • Other duties as assigned

REQUIRED QUALIFICATIONS
  • Bachelor's degree with 8+ years, Masters with 6+ years, or PhD with 3+ years experience in Molecular Biology or related area
  • Background in high throughput molecular biology techniques, e.g. DNA builds
  • Experience with automation
  • Experience with analysis of large datasets

PREFERRED QUALIFICATIONS
  • Industry experience in molecular biology techniques for protein engineering/evolution strongly preferred
  • Familiarity with liquid handlers such as Hamilton/Tecan/ECHO
  • Familiarity with colony pickers such as Qpix
  • Experience with plate readers
  • Experience with CRISPR systems
  • Computer and scripting skills (Benchling, Geneious, JMP, Python)

TEAM
Our small-size proteins, such as Cas14 and CasPhi, are poised to transform disease treatment enabling novel editing approaches and delivery options that are difficult to achieve or not even possible with previously described CRISPR-Cas systems. Our teams are responsible for discovering and engineering novel CRISPR systems, establishing CRISPR-based diagnostics, and translating our unique CRISPR technology into therapeutic treatments with the ultimate goal of cures for indications with high medical need.
