Job Description

The Vector Engineering laboratory led by Ben Deverman within the Stanley Center for Psychiatric Research at the Broad Institute of Harvard and MIT is looking for self-motivated research associates with experience producing and purifying viral vectors and performing quality control assays. The positions provide the opportunity to join a mission driven team focused on expanding the impact of gene therapy by developing next generation adeno-associated viruses (AAV). The successful candidate will work closely with members of the Vector Engineering team to support AAV production and purification, high-throughput AAV screening, and the development and implementation of quality control assays and standard operating procedures. Successful candidates will also contribute to collaborations with other Broad Institute scientists and teams, and will be expected to function independently in his/her daily responsibilities after training.


* Contribute to the design, implementation and troubleshooting of experiments critical to the team's collective success

* Maintain adherent and suspension cell lines and perform AAV production, downstream processing, quantification and quality control assays

* Design and perform molecular biology experiments such as plasmid construction, sequencing and QC assays, quantitative PCR, digital droplet PCR, RNA, DNA and protein extractions from cell or tissue samples, next generation sequencing (NGS) library preparation

* Perform immunohistochemistry, reporter assays and FACS

* Consult colleagues and the scientific literature, as needed, to support ongoing and future projects

* Maintain meticulous records and document, compile, and analyze experimental data

* Assist with laboratory organization, ordering, and the operation and maintenance of laboratory equipment, and perform other duties as assigned

* Attend and contribute to team meetings

* Make contributions to scientific manuscripts and conference presentations


* B.S. or M.S. with 3+ years of cell culture experience in industry or academic vector core

* Hands-on experience and extensive knowledge of mammalian cell culture, transfection, and recombinant protein or virus production

* Upstream and/or downstream process development for biologics (AAVs, vaccines, mAb, or recombinant proteins)

* Experience performing a variety of analytical assays

* Exceptional organizational and time management skills, lab notebook practices and careful attention to detail

* Outstanding interpersonal and communication skills

Hands on experience with any of the following is a plus:

* Stable cell line generation and clonal analysis

* CRISPR/Cas9 and/or siRNA-based gene knockout / knockdown

* Experience with common molecular biology and cloning techniques (RNA/DNA isolation, restriction-ligation and assembly based cloning, DNA sequencing and construct design).
