Position description

A postdoctoral position is available at the University of California, Merced to study the molecular mechanisms for DNA recognition, scanning and nucleosome engagement by pioneer transcription factors and their role in cell fate decisions. The project addresses the investigation of DNA recognition processes by pioneer transcription factors that function as master regulators of cell fate with a focus on the mechanisms of DNA protein interactions, DNA scanning, protein conformation and DNA shape changes induced by binding, as well as their

mechanical effects on chromatin. The research will primarily involve protein biochemistry combined with advanced single-molecule biophysical methods, including ultrafast smFRET spectroscopy, force spectroscopy via optical tweezers, and correlative confocal fluorescence microscopy to investigate protein-DNA interaction dynamics. Experimental biophysics studies will be complemented with computer simulations and high-throughput DNA-binding selection assays and genome editing studies of Candida albicans as model system to investigate the significance of the mechanisms we identify biophysically in controlling a simple in vivo model of cell fate.

The successful candidate will be part of a team of postdoctoral, graduate, and undergraduate students that is one of the world leaders in protein biophysics research (more information at http://vmunoz-group.ucmerced.edu/). The lab is currently embarked in multiple basic research projects that aim at understanding a variety of biological problems related to the conformational dynamics of proteins, including, the molecular biophysics of the control of gene expression, the experimental and theoretical investigation of the mechanisms of protein folding through the detection of folding transition paths (e.g. see https://pubs.acs.org/doi/full/10.1021/acs.accounts.0c00322), and the analysis of folding coupled to binding of intrinsically disordered proteins (e.g. see https://www.pnas.org/doi/abs/10.1073/pnas.2113572119). In addition, the lab is embarked in the development of new protein-based technologies including genetically encoded high-performance fluorescence biosensors, allosterically controlled macromolecular assemblies, and protein decoy fluorescence diagnostics methods.

The postdoctoral researcher is expected to stay abreast of recent publications in the field, carry out research independently, implement and develop research ideas and write research articles. The postdoctoral scholar is also expected to assist with mentoring graduate students and undergraduate students who will work on the project, and help in properly maintaining the spectroscopic and optical equipment in the lab. This is a two-year appointment will possible extension based on satisfactory performance.

Qualifications

Basic qualifications
PhD, MD (or foreign equivalent) in Bioengineering or related field.

Excellent organization, writing, presentation, and communication skills.

Ability to conduct independent research.

Preferred qualifications
Previous training and established track record in experimental protein biophysics and/or biochemistry/protein chemistry.
