Job Purpose

We seek to recruit a full-time postdoctoral-level Research Fellow in Sequencing Molecular Biology to work in the laboratory of Professor Nick Loman on an exciting Wellcome Trust Technology Development Award to develop new methods for long read sequencing to enable - for the first time - complete whole genome assemblies of complex genomes.

Working with an experienced team and in close collaboration with Professor Matt Loose's laboratory at the University of Nottingham we are developing an collaborative, worldwide, open approach to developing new molecular biology methods for use on long-read sequencing platforms (e.g. Oxford Nanopore, Pacific Biosciences, 10X Genomics, BioNano, etc.). Your work will be part of our newly formed Long Read Club ( longreadclub.org ). The grant aims to enable end-to-end chromosomal-level assemblies (including telomeres and centromeres) routinely at scale for genome sequencing projects.

The Research Fellow in Sequencing Molecular Biology will focus primarily on developing new methods for long fragment DNA extraction and molecular biology methods for sequencing library construction in order to reliably generate multi-megabase length sequencing reads, going beyond kits. This will include development and refinement of existing transposase-based protocols (e.g. our existing ultra-long read protocol: https://www.protocols.io/view/ultra-long-read-sequencing-protocol-for-rad004-mrxc57n ), new methods to protect high molecular weight DNA during library preparation and rediscovery and testing of forgotten extraction and molecular biology techniques. You will also take part-responsibility for Long Read Club protocols and online meetings to widely disseminate the outputs of the grant and to build a community of users interested in these methods.

The project will offer ample opportunity for authoring primary molecular biology research outputs and support for further career development in academia, e.g. fellowship applications. You will work in a lab with extensive molecular biology experience (working with Dr Josh Quick) and bioinformatics expertise.

Person Specification
  • Applicants should hold a PhD in biology, molecular biology, genomics, sequencing or chemistry, with some experience in the development of molecular biology methods and protocols.
